  • Palbociclib is mainly used to manage hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative advanced or metastatic breast cancer. It belongs to a class of medications known as cyclin-dependent kinase (CDK) 4/6 inhibitors.

  • It works by interrupting the process of cell division in cancer cells. Specifically, it blocks the action of proteins known as cyclin-dependent kinases 4 and 6. These proteins are involved in cell cycle progression, which controls cell growth and division. By inhibiting their action, Palbociclib slows down the rapid cell division seen in cancer cells, thereby slowing the growth of tumours. Palbociclib has also been found to inhibit a cellular process called Rb phosphorylation, which is another critical process involved in allowing cells to progress from one stage of cell division to the next. Ultimately, through these actions, Palbociclib helps in managing the spread and progression of certain types of advanced or metastatic breast cancers.
